In a recent paper, we showed that human subcutaneous mature adipose cells assume a senescent phenotype in obese individuals, and this was even more pronounced in T2D individuals independent of BMI and age (Gustafson et al. 2022) (Fig. 2).γH2AX staining, as a measure of unresolved DNA damage response, the senescence marker CDKN1A as well as the expression of SASP factors (IL6, CCL2, SERPINE1, and IL1B) were increased in mature adipose cells from obese hyperinsulinemic individuals (Li et al. 2021). Here, SERPINE1 is linked to type 2 diabetes mellitus.
