In order to further understand why the results in human cancer cells are inconsistent with those in mice, and further clarify the role of ZKSCAN3 in the transcriptional regulation of autophagy in human cancer and non-cancer cells, we used the CRISPR/Cas9 system to knock out the ZKSCAN3 gene in human HK-2 cells (non-cancer cells) and HeLa cells (cancer cells) [7,8], and constructed monoclonal HK-2 cell lines lacking ZKSCAN3 and monoclonal HeLa cell lines lacking ZKSCAN3, respectively.
