The KEYNOTE-189 study 26 compared the efficacy and safety of pembrolizumab combined with pemetrexed and platinum versus placebo combined with pemetrexed and platinum in treatment-naïve EGFR or ALK-negative metastatic non-squamous NSCLC patients, with KRAS mutations in 89 (31%) patients, and pembrolizumab combined with platinum-based doublet chemotherapy showed more PFS benefit compared with platinum-based doublet chemotherapy (mPFS 9.0 and 5.0 months, HR = 0.47, 95% CI: 0.29 to 0.77), with a similar trend of OS benefit (mOS 21.0 and 14.0 months, HR = 0.79, 95% CI: 0.45 to 1.38). The gene discussed is ALK; the disease is non-small cell lung carcinoma.
