In subcutaneous xenograft models of resistant PDX lines, GNPAT deletion alone did not impede tumor growth; however, treatment of animals harboring GNPAT-deleted tumors with IACS-010759 significantly suppressed tumor growth and increased cell death (Fig. 4f, g, Supplementary Fig. 17a, b), underscoring the importance of ether lipids in resistance to complex I inhibition.
