A recent study showed that HIF-1α stabilized by myriocin, an inhibitor of the de novo synthesis of sphingolipid, could promote the expression of PDK1 (3-phosphoinositide-dependent protein kinase-1, an enzyme involved in glycolysis) and BNIP3 (BCL2/adenovirus E1B 19 kDa protein-interacting protein 3, a member of the apoptotic Bcl-2 protein family) and alter the intracellular levels of glucose metabolites, to inhibit erastin- or glutamate-induced ferroptosis in HT-1080 human fibrosarcoma cells, but not in GES-1 human gastric epithelial cells and SK-Hep-1 human hepatoma cells [109]. The gene discussed is BCL2; the disease is fibrosarcoma.
