Caplacizumab (also known as ALX-0081) is a single-domain nanobody that inhibits the vWF-GPIb/IX/V interaction by blocking the vWF A1 domain [109] In Phase I studies in healthy subjects and stable angina patients undergoing percutaneous coronary intervention (PCI), Caplacizumab was well tolerated and effectively inhibited pharmacodynamic markers [110] Unfortunately, further studies did not prove the effectivity of Caplacizumab in patients with high-risk PCI, however this approach was successfully used in the treatment of thrombocytopenic purpura [111].
