Interestingly, our CD4+ T cells model predicts that in response to high insulin levels, regulatory T cells are polarized toward IL-10-producing lineages like Tr1 to the detriment of TGFβ-producing lineages such as Th3 (Figure 6G), which is consistent with experimental observations, in which it can be verified that one of the physiological adaptations of obesity is the production of IL-10 (43) and the decrease in TGFβ is associated with the appearance of insulin resistance in obese persons (42). The gene discussed is TGFB1; the disease is obesity due to melanocortin 4 receptor deficiency.
