Recent research published in Nature Neuroscience has revealed that lysosome dysfunction that causes autophagy failure has a close correlation with AD pathology and Aβ deposition, that is, autolysosome acidification declines in neurons before extracellular amyloid deposition, associated with a significant decrease in v-ATPase activity and selective accumulation of Aβ/Aβ precursor protein (APP) within enlarged de-acidified autolysosomes 105.
