A flow cytometry-based assay was used to measure the accumulation of cytotoxic granule membrane protein, LAMP1/CD107a, at the surface of NK cells (as a proxy for granule release); while simultaneously detecting production of interferon γ (IFNγ) on a per NK cell basis after PBMC were co-incubated with these NSCLC lines for 5 h (Figures 5A, B, S4).
