Roulin et al. (2011) revealed that combined treatment with sorafenib and NVP-BEZ235, a novel dual PI3K/mTOR inhibitor, demonstrated enhanced antitumor efficacy in RCC cell lines, 786-0 and Caki-1, compared to either of the single treatments. Other mTOR inhibitors, such as mTORC2 inhibitors, PP242 and PP30 (Feldman et al., 2009; Li et al., 2021), and the mTORC1/2 dual inhibitor, WYE-125132 (Yu et al., 2010), all demonstrated ideal antitumor capacity in RCC preclinically.
