PRKN and Parkinson disease: Both Parkin and VPS35 are implicated in the pathogenesis of Parkinson's disease (Panicker et al., 2021), and, although the mechanistic consequences of their interplay remain unclear (Williams et al., 2017), Parkin appears to promote retromer complex integrity, as brains of Parkin-knockout mice show marked depletion of retromer-associated WASH proteins (Williams et al., 2018).
