Patatin-like phospholipase structural domain 3 (PNPLA3) is the major susceptibility gene for NAFLD, and its PNPLA3 I148M (rs738409, C > G) is strongly associated not only with hepatic fat content [2] but also with the progression of NAFLD to NASH, fibrosis, and cirrhosis [3]. This evidence concerns the gene PNPLA3 and metabolic dysfunction-associated steatohepatitis.
