BRAF and colorectal carcinoma: Oncogenic mutations in BRAF, a potent modulator of the MAPK pathway present in approximately 10% of CRC patients6,7, can now be successfully targeted with the combination of encorafenib (BRAF inhibitor) and cetuximab, which are now standard-of-care after the positive BEACON trial in 2019, and/or dabrafenib (BRAF inhibitor) and trametinib (MEK inhibitor) which were both introduced in 20148–11.
