TP53 and cancer: Mutations in the p53 gene occur in a wide array of human cancers (www-p53.iarc.fr, accessed on 12 July 2022), predominantly through missense mutations localized to six “hotspot” residues which can be classified as either contact mutants (which disrupt DNA binding activity, e.g., R273H) or structural mutants (hallmarked by altered conformation, e.g., R175H).
