In early clinical trials, adding EGFR-TKIs to chemotherapy showed no significant improvement of PFS, partially because patients were not selected for EGFR-sensitive mutation.13,14,15 However, clinical trials showed that the combination of chemotherapy and gefitinib could significantly improve PFS in patients with EGFR-mutant NSCLC, which makes a case for revisiting the combination therapy strategy.16,17,18 The NEJ009 Study demonstrated superior PFS benefit in a subgroup analysis of patients with brain metastases,17supplying a promising strategy for these patients. The gene discussed is EGFR; the disease is non-small cell lung carcinoma.
