Based on the observations in lung cancer (Awad et al., 2021; Zhao et al., 2021) and on predictions based on mouse models that allow genetic inactivation of oncogenic KRAS (Collins et al., 2012; Ying et al., 2012), it is to be expected that resistance to KRAS G12D inhibitors will similarly arise; activation of anti-tumor immune responses remains our best hope for long-term tumor control.
