The key enzymes of glycolysis, including hexokinase 2 (HK2), 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase-3 (PFKFB3) and pyruvate kinase M2 (PKM2), promote the conversion of glucose to lactate instead of pyruvate in the tumor cells, which in turn impairs mitochondrial oxidative phosphorylation (Minchenko et al., 2002; Yang et al., 2014; Xu and Herschman, 2019).
