Clinical studies have found that the low-density lipoprotein receptor-related protein 5 (LRP5), a key component of canonical Wnt/β-catenin pathway, is critical to osteogenesis, with a loss-of-function mutation impeding bone development to cause osteoporosis-pseudoglioma syndrome11 and a gain-of-function mutation increasing bone mass.12 The gene discussed is LRP5; the disease is osteoporosis.
