It is important to note that the high expression of GADD34 correlates with a shorter duration of response to anti-Her2 therapy in breast cancer patients in the clinic (Fig 5H), suggesting that our findings from our in vitro model reveal clinically relevant mechanisms, and that targeting of the ER quality control system might be a viable therapeutic option post-progression on Her2 inhibitors.
