Vemurafenib, a BRAF kinase inhibitor, was approved by the FDA for metastatic melanoma with a BRAF V600E mutation in 2011 based on results of a phase 3 BRIM-3 trial comparing vemurafenib against dacarbazine.22 Overall survival was 84% (95% CI, 78%-89%) with vemurafenib and 64% (95% CI, 56%-73%) with dacarbazine.
