By age 70 years, pathogenic variants in MLH1 and MSH2 convey a 46 and 35% risk of CRC development, respectively, despite colonoscopic surveillance, whereas those with pathogenic variants in MSH6 or PMS2 observe markedly lower cumulative incidence rates by the same age: MSH6 (20%) and PMS2 (0-1%) [2]. This evidence concerns the gene MSH6 and colorectal carcinoma.
