To determine if the variant-based changes in BAFF-induced AKT phosphorylation detected in the DG-75 model would correlate with changes in the phosphorylation of S6 in primary B cells, we activated PBMCs of CVID patients carrying P21R, G64V, DUP92-95, and H159Y with BAFF and analyzed pS6 (S240/244) by intracellular flow cytometry. The gene discussed is TNFSF13B; the disease is common variable immunodeficiency.
