GMB-475 (Burslem et al., 2019), targeting the myristoyl pocket of ABL1 via an allosteric way, degrades the BCR::ABL1 fusion protein through the ubiquitin–proteasome pathway; however, it inhibited proliferation and promoted apoptosis in CML cell lines only at high concentrations. The gene discussed is ABL1; the disease is chronic myelogenous leukemia, BCR-ABL1 positive.
