These critical knowledge gaps are partly due to the complexity of tau: there are 6 isoforms of tau in the human brain, all of which differently accumulate in tauopathies (e.g. aggregates of 3R tau isoforms accumulate in Pick’s disease, aggregates of 4R tau isoforms accumulate in PSP and CBD [37].
