ZBP1 and cancer: Hanahan (2022) suggested that resistance to cell death is one of the basic hallmarks of cancer. Caspase-8 (CASP8) is a molecular switch that controls pyroptosis, apoptosis, and necroptosis (Fritsch et al., 2019). Jiang et al. reported that CASP8 is a key protein in the crosstalk signaling pathway of PANoptosis in cancer (Jiang et al., 2021). A recent study (Karki et al., 2021a) showed that ZBP1 activates PANoptosis through RIPK3 signaling, ADAR1 negatively regulates ZBP1-mediated apoptosis, and blocking ADAR1 activity contributes to apoptosis and inhibits tumorigenesis.
