To verify that the regulation of autophagy and the improvement of renal fibrosis by HDHW are related to the IGF-1-mediated PI3K/Akt/mTOR signaling pathway, this study blocked the binding of IGF-1 to IGF-1R by intraperitoneal injection of IGF-1R blocker JB1 and observed the expression of downstream signaling factors, including PI3K, Akt, mTOR, p-PI3K, p Akt, p-mTOR, as well as the expression of autophagy marker proteins including Beclin1, LC3II/I, and renal fibrosis marker proteins including α-SMA. This evidence concerns the gene AKT1 and renal fibrosis.
