Using in vitro infection models with BCG and Salmonella, myeloid lineage THP-1 cells generated using shRNA to achieve partial JAK1-deficiency (25-30% residual JAK1 protein expression) supported enhanced bacterial survival after IFN-γ stimulation, reminiscent of uncontrolled mycobacterial replication previously reported in IFN-γR1-deficient human iPSC-derived macrophages (51).
