Drug screening revealed that ponatinib, an FDA-approved multi-kinase inhibitor for leukemia, was effective against RET G810S, which was confirmed by analyses of the crystal structure of RET using computer-aided docking poses of truncated analogs of selpercatinib and ponatinib: the resulting serine alcohol in RET G810S clashes with selpercatinib but not with ponatinib (Fig. 7b–d and Supplementary Fig. 6f). The gene discussed is RET; the disease is leukemia.
