PNLIP and non-small cell lung carcinoma: While these studies pointed out the potential pharmaceutical value of targeting EGFR palmitoylation for treating NSCLC, interestingly, another group presented that blocking FASN with Orlistat (a reversible inhibitor of gastric and pancreatic lipases) treatment prevents EGFR palmitoylation, enhances ubiquitination and degradation of EGFR, and then inhibits NSCLC cell proliferation, and suppresses in vivo tumorigenesis [85] (Fig. 2, Table 1).
