The link between protein palmitoylation and NSCLC was established by the finding that ZDHHC21 palmitoylates EGFR at Cys1025, Cys1034, and Cys1122 in HEK293T cells, where EGFR and ZDHHC21 were coexpressed and purified for mass spectrometry; and inhibition of ZDHHC21 with shRNA in MDA‐MB‐231 cells or expressing the palmitoylation site mutation of EGFR plasmids in NIH 3T3 cells enhanced EGFR activation and increased EGFR signaling‐dependent cell survival and migration [83].
