Overexpression of wild-type BAP1 in lung cancer NCI-H226 cells significantly inhibits the tumorigenic ability of the cells in nude mice, while overexpression of BAP1 with either enzyme-activity mutation (C91A) or nuclear localization sequence deletion (NLS2-Ala) did not affect the tumorigenic ability (29), suggesting that the anti-tumor function of BAP1 depends on its catalytic activity and nuclear localization.
