Among Cas9D- and Cas9H-induced GFP−RFP+ cells, the proportions of INV3, a product of MH-mediated TD, were increased from none in both in Brca1+/+ cells to 32.5% and 11.8% in Brca1m/m cells, respectively, whereas Brca1 deficiency stimulated nCas9-induced INV1 and INV2, a product of TRSI-mediated TD, to a lesser extent (i.e., Cas9H-induced INV1) or not at all (i.e., Cas9H-induced INV1 and INV2) (Fig. 6b). Here, BRCA1 is linked to thanatophoric dysplasia.
