We focused on p53 to understand the mechanism of SIRT1 down-regulation in post-RLDC kidneys mainly based on two facts: 1) RLDC induced a significant decrease in SIRT1 mRNA in BUMPT cells indicating SIRT1 down-regulation at the transcription level (Figure 7E), and p53 has been reported to regulate SIRT1 transcription (46); and 2) p53 is known to mediate cisplatin-induced AKI (47, 48), and we detected p53 activation in post-RLDC mouse kidneys (Figure 8E). This evidence concerns the gene SIRT1 and acute kidney injury.
