Therefore, we engineered adenovirus-associated virus (AAV) with a soluble form (sTRAIL, aa 114–281) and IFNB1 fused with an isoleucine zipper (ILZ) for trimerization, a signal peptide of the human fibrillin gene that ensures effective secretion and a furin cleavage site to release the ILZ-sTRAIL protein into the extracellular space and a P2A cleavage site to maintain IFNB1 in the cytoplasm under the control of the CRC-specific CEA promoter (Fig. 5A).
