In human CNS, nAChRs are pentamers composed of various combinations of twelve different subunits (α2–α10 and β2–β4), with the homo-pentameric α7-nAChR (all α7 subunit) and hetero-pentameric α4β2-nAChR (α4 and β2 subunits in a 2 : 3 or 3 : 2 ratio) being predominant.1 α7-nAChR is mainly expressed in the hippocampus and cortex regions which are involved in cognition and memory.2 Clinical studies have demonstrated that α7-nAChR is implicated in a variety of neurodegenerative and neuropsychiatric disorders such as Alzheimer's disease (AD) and schizophrenia. Here, CHRNA7 is linked to early-onset autosomal dominant Alzheimer disease.
