To do this, plasmids encoding wildtype (WT) or Mafa-4A were transfected into a human neuronal cell lines (medulloblastoma, DAOY) that do not express MAFA. As measured by RT-qPCR, Mafa significantly increased GAD2 expression in both cell lines (Fig. 5b) with GAD2 expression being four times higher in Mafa-4A expressing cells. Here, MAFA is linked to medulloblastoma.
