SIK2 and ovarian cancer: In collaboration with Arrien Pharmaceuticals, we have developed orally administered low-MW drugs (ARN3236 and ARN3261) that inhibit SIK2 at nanomolar concentrations, inhibit growth of ovarian cancer cell lines with an IC50 of 0.8 to 2.6 μM, and inhibit growth of ovarian cancer xenografts, enhancing sensitivity to paclitaxel (18) and carboplatin (19).
