Co-immunoprecipitation (CoIP) studies with human hepatoblastoma cells (HepG2 cells) and GST-pull down assays performed using human embryonic kidney cells (HEK293T cells) revealed that the binding of PROX1 to the promoter represses the expression of target genes by recruiting the LSD1/NuRD complex, directly binding to the LSD1 unit, and promoting H3K4 hypomethylation. The gene discussed is KDM1A; the disease is hepatoblastoma.
