PARP1 and acute myeloid leukemia: The recently published SOLO-1 trial evaluating frontline olaparib maintenance also showed a 1% incidence of AML in the treatment arm (compared to 0% in the placebo arm), which is worrisome because it suggests that AML may be a toxicity specific to PARP inhibitor treatment, and not related to prior chemotherapy, since these were patients treated in the frontline setting[27].
