Furthermore, to test how TFAP2 paralogs affect enhancer activity and MITF binding, we deleted TFAP2 paralogs from SK-MEL-28 melanoma cells and assessed: nucleosome positioning, using the assay for transposase-accessible chromatin with next generation sequencing (ATAC-seq); enhancer activity, using cleavage under targets and release using nuclease (CUT&RUN) with anti-H3K27Ac, anti-H3K4Me3, and anti-H3K27me3; and binding of MITF, using CUT&RUN with anti-MITF.
