Based on this finding and the fact that MITF-M isoform, which is the type of isoform expressed in melanomas, lacks this N-terminal domain which is required for lysosomal localization and mTOR phosphorylation, it is proposed that overexpressing MITF-M binds promiscuously to CLEAR element lysosomal genes without being restrained by mTOR signaling and hence regulates endolysosomal biogenesis [25].
