PSEN1 and autosomal dominant disease: EOAD is an autosomal dominant disease with heritability of more than 70% (Gatz et al., 2006; Wingo et al., 2012) and three responsible mutated genes, the amyloid protein precursor gene (APP), presenilin-1 gene (PSEN1), and presenilin-2 gene (PSEN2), were found to mainly dominate the production, aggregation, and clearance of amyloid β-protein (Aβ) (Cacace et al., 2016).
