Small molecule inhibitors of TEAD autopalmitoylation have recently been demonstrated to bind TEAD proteins and prevent TEAD autopalmitoylation, disrupt YAP/TAZ interaction with TEAD proteins, diminish YAP/TAZ:TEAD transcriptional co-activation, downregulate YAP/TAZ:TEAD target transcripts, and inhibit in vivo tumor growth of NF2-deficient cancer cell lines (Figure 4) [123].
