While this study has some limitations, it provides the first evidence that a targeted approach using site-specific demethylation and transcriptional activation, is a feasible method of restoring LRIG1. As LRIG1 is known to play a crucial role in tumorigenesis and metastasis, combining CRISPR/dCas9-based reactivation with conventional therapeutic approaches could hold promise for LRIG1-silenced tumours.
