We found that the decline in FAM134B was due to lysosomal degradation as inhibition of lysosomal activity using concanamycin A prevented FAM134B degradation during HE-hypoxia (Fig. 2g and Supplementary Fig. 2j) and CI-hypoxia (Supplementary Fig. 2k). The gene discussed is RETREG1; the disease is hereditary elliptocytosis.
