Recent studies found that the expression of some pyroptotic inflammasomes and gasdermins decreased in cancer cells.14,15 Drug-regulated pyroptosis promotes inflammatory cell death of cancer and inhibits proliferation and migration of cancer cells.16,17 Val-boroPro, the ‘inflammasome’ sensor protein CARD8 activator, which successively activates procaspase-1 to mediate pyroptosis in primary acute myeloid leukemia (AML) samples and most AML cell lines,18 suggesting that pyroptosis is suitable for the treatment of AML. This evidence concerns the gene CARD8 and cancer.
