In order to evaluate whether intrauterine hyperinsulinemia induced the abnormal expression of FoxO1 and DNMT3A, we cultured HepG2 in different insulin concentrations, and the results indicated that the expression of FoxO1 and DNMT3A gradually augmented as the insulin concentration increased, at both the mRNA and protein levels (Figures 5B–D). This evidence concerns the gene INS and hyperinsulinism.
