Next generation anti-CD33s are under development, including vadastuximab talirine (SNG33A), a humanized murine anti-CD33 IgG1 mAb coupled with two molecules of pyrrolobenzodiazepine, which showed good tolerability in monotherapy and an encouraging CCR (CR + CRi 73%) in a study on 53 elderly AML patients who were ineligible for IC [98,99].
