A preclinical/clinical phase II study demonstrated that bortezomib can selectively sensitize patients with relapsed/refractory ABC-DLBCL, but not patients with relapsed/refractory GCB-DLBCL, to chemotherapy.31 Furthermore, bortezomib has limited single-agent activity in relapsed/refractory DLBCL.32 Another example is small-molecule BH3 mimetics which are BCL2 inhibitors, including ABT-737, ABT 263, and GX15-070. The gene discussed is BCL2; the disease is diffuse large B-cell lymphoma.
