Covalent inhibitors of KRAS G12C have demonstrated response rates of 32% to 45% in NSCLC with activity observed in other tumor types as well (eg, pancreatic, cholangiocarcinoma, endometrial, and ovarian cancers).34,35 Recently, the KRAS G12C inhibitor sotorasib was approved for locally advanced or metastatic KRAS G12C-mutated non-small-cell lung cancer patients who have received at least one prior systemic therapy. Here, KRAS is linked to non-small cell lung carcinoma.
