When looking at essentiality from published genome-wide CRISPR screens of AR-regulated genes in PCa cells (LNCaP) (Figure 1), we find many known and novel essential genes, including coactivators such as GRHL2 (grainyhead-like transcription factor 2) [25]; metabolic genes such as DNM1L (dynamin-related protein 1) [26], SREBP (sterol regulatory element-binding protein) cleavage activating protein SCAP [27] and mTOR (the protein kinase mammalian target of rapamycin) [28]; and transcriptional regulators such as NFKBIA (NFKB inhibitor alpha), an inhibitor protein of NF-κB and p53 [29]. Here, MTOR is linked to posterior cortical atrophy.
